背景
成纖維細(xì)胞生長(zhǎng)因子10(FGF10)是成纖維細(xì)胞生長(zhǎng)因子家族(FGF)的一員,屬于FGF7亞家族,與FGF3、FGF7、FGF22等成員具有結(jié)構(gòu)相似性。是人體皮下的組織細(xì)胞分泌的一種堿性蛋白生長(zhǎng)因子,能特異刺激上皮細(xì)胞的新陳代謝等生理過程,包括細(xì)胞的再生、分化和遷移等。
在胚胎期調(diào)控肺、乳腺、前列腺、唾液腺等器官的形態(tài)發(fā)生,例如促進(jìn)肺芽分支和腺體形成。能夠激活MAPK通路,促進(jìn)細(xì)胞增殖和遷移;通過PI3K/Akt通路抑制細(xì)胞凋亡,增強(qiáng)細(xì)胞存活;與肝素結(jié)合蛋白多糖(HSPG)、Klotho蛋白等輔因子結(jié)合,增強(qiáng)信號(hào)傳遞效率。
成纖維細(xì)胞生長(zhǎng)因子10有很好的臨床應(yīng)用前景,在慢性創(chuàng)面,如糖尿病潰瘍,通過促進(jìn)血管新生和上皮再生加速愈合;在帕金森病、阿爾茨海默病中促進(jìn)神經(jīng)元存活;部分FGF家族成員與腫瘤發(fā)生相關(guān),F(xiàn)GF10的靶向調(diào)控可能成為治療策略。
